Tato homologn\u00ed sekvence lemuje sekvenci DNA st\u00e1vaj\u00edc\u00edho genu jak p\u0159ed, tak i za um\u00edst\u011bn\u00edm va\u0161eho genu na chromozomu. Atomov\u00e9 mechanismy nov\u00e9 bu\u0148ky automaticky rozpozn\u00e1vaj\u00ed stejn\u00e9 \u00faseky sekvence a nahrazuj\u00ed st\u00e1vaj\u00edc\u00ed gen nebo jeho \u010d\u00e1st fale\u0161nou \u010d\u00e1st\u00ed DNA. Proto\u017ee fale\u0161n\u00e1 DNA je mrtv\u00e1, ovliv\u0148uje pouze d\u011bdi\u010dn\u00fd gen, jinak \u0159e\u010deno \u201ereport\u00e9rov\u00fd gen\u201c, dostupn\u00fd pro pou\u017eit\u00ed v z\u00e1znamu, nov\u00e1 v\u00fdm\u011bna odstra\u0148uje nebo \u201evy\u0159azuje\u201c \u010d\u00e1st st\u00e1vaj\u00edc\u00edho genu. Ve t\u0159ech nez\u00e1visl\u00fdch testech jsem elektroporoval \u010derstvou linii bun\u011bk K562 BCR\/ABL, kter\u00e1 obsahuje SDE-hABL-1 a browser-hABL-1sgRNA.<\/p>\n<p>Knockout, jak se vztahuje k genomice, ozna\u010duje pou\u017eit\u00ed genetick\u00fdch technologi\u00ed k inaktivaci nebo ztr\u00e1t\u011b alespo\u0148 jednoho specifick\u00e9ho genu ze syst\u00e9mu. Nakonec jsem sekvenoval v\u0161echny vytvo\u0159en\u00e9 mutantn\u00ed alely a mohl jsem analyzovat d\u016fsledky in silico i in vivo.
